Potensi Senyawa Nitazoxanide dan Arbidol sebagai Antivirus SARS-CoV-2 terhadap Reseptor NSP5 (7BQY dan 2GZ7) dan ACE2 (3D0G dan 1R4L)

چکیده

Nitazoxanide merupakan obat pro-drug untuk tizoxanide. Obat tersebut diketahui memiliki aktivitas antivirus pada MERS-CoV. Arbidol H3N2 HA yang kesamaan struktur dengan SARS-CoV-2. Reseptor target spesifik obat-obat Anti SARS-CoV-2 saat ini adalah NSP5 (Nonstructural protein) dan ACE2 (Angiotensin-converting enzyme 2). memediasi replikasi transkripsi protein-protein virus sedangkan sebagai fasilitator transport asam amino reseptor tempat terikatnya SARS-CoV maupun Tujuan penelitian mengetahui dari senyawa secara in silico. Senyawa dipreparasi terlebih dahulu melakukan pengunduhan kode PDB ID, penyiapan 2D 3D, penentuan minimal energy, pembuatan SMILES serta prediksi sifat fisikokimia toksisitas. Setelah dipreparasi, dilanjutkan proses penambatan molekul menggunakan aplikasi PyRx-Vina®. Hasil docking dianalisis perangkat lunak PyMOL® Discovery Studio Visualizer®. profil menunjukkan bahwa telah memenuhi hukum Lima Lipinski masing-masing nilai LD50 sebesar 1350 mg/kg 340 mg/kg. uji menghasilkan interaksi yaitu ikatan hidrogen, hidrofobik elektrostatik. Interaksi 2GZ7 3D0G 1R4L perbedaan signifikan terhadap Lopinavir Chloroquine p < 0.05, sehingga diprediksi potensi lebih baik.
